Bacteria Lab
Step 1: Obtain proper safety gear
Step 2: Check your equipment
Step 3: Obtain 2 pieces of aspirin from Ms. Lyman
Step 4: Grind the two pieces using the mortar and pestle on the back table
Wipe the powder into your beaker
CAREFUL NOT TO SPILL, TRY TO GET ALL OF TEH POWDER INTO THE BEAKER
Step 5: Get 5mL of methanol from the fume hood
Pour this in your beaker and stir the mixture until it is cloudy
The pill will NOT dissolve completely
DO NOT BREATHE OVER THE BEAKER
Step 6: Fold your filter paper into the funnel
Place funnel/ paper in tube
Put your tube/ funnel set in rack and pour the solution through the filter into the tube
Write your group's initials in sharpie top to stop
Step 7: Go to fume hood, Ms. Lyman will add 3 drops of sulfuric acid (H2SO4) to the tube and place on a hot water bath for 30 minutes
Step 8: Make an observation on your new material
Transfer to vial using your pipette
You may need to pour it through the beaker to make this easier

- small beaker
- 2 small paper towels
- test tube
- test tube rack

Determine the 'mintiest' smelling sample
Step 3: Get a petri dish with ager from Ms. Lyman
Write the names of your group (2-3 people) along the outside of the petri dish.
Step 4: Acquire samples of distilled H2O and methanol (control)
Always want a positive and negative control
Step 5: Acquire a sample of acetyl acid for your before sample (aspirin)
Step 6: Place one 'dot' of filter paper in each of your labeled trays
Place 6 drops of that sample type on the dot
Step 7: Label your plates + and - as follows
+ positive control *methanol*
- negative control *H2O*

Step 8: Use a sterile loop to pick up a single bacterial colony form the starter plate
Spread the bacteria on your plate in a star pattern while rotating the plate until the surface is covered in bacteria
DO NOT GOUGE AGAR
Step 9: Place the correct dot on top of the agar in the correct dish
DOUBLE CHECK
Step 10: Cover and wrap in para film
Place on back tray for correct class
Step 11: Place loop in bleach bucket to sanitize
Place gloves in designated HAZARDOUS WASTE CONTAINER
Step 12: Wash hands
Step 13: Put away materials on lab trays
Return goggle to lab
Spread the bacteria on your plate in a star pattern while rotating the plate until the surface is covered in bacteria
DO NOT GOUGE AGAR
Step 9: Place the correct dot on top of the agar in the correct dish
DOUBLE CHECK
Step 10: Cover and wrap in para film
Place on back tray for correct class
Step 11: Place loop in bleach bucket to sanitize
Place gloves in designated HAZARDOUS WASTE CONTAINER
Step 12: Wash hands
Step 13: Put away materials on lab trays
